Basically I want to use uniform server as a testing site for changes which I am able to do now perfectly and also have it work as a backup server if my regular server should crash. I need to know how to set virtual server so that in case of emergency and I point the dns to my local computer it resolves to www.uslister.com when the directory is now http://localhost/joomla/. Also how can I set it to use my mail provider optonline.net to send the emails? Joomla has email settings in admin panel but I was unsuccesful in setting it up.

For the mail, try entering the SMTP information into the php.ini file, and about the backup use.. search the forum for post on virtual host, dynamic dns... and also search Google....
